Creative Biolabs Unveils Dual-Vesicle Platform to Tackle CAR-T Barriers in Solid Tumors

Creative Biolabs has introduced an integrated OMV and exosome-boosted CAR-T development platform aimed at investigating strategies to overcome the barriers that have constrained CAR-T cell therapy in solid-tumor models. The announcement, made by the global biotechnology and contract research services provider, centers on a next-generation vesicle-engineering approach that combines bacterial outer membrane vesicles (OMVs) and mammalian-derived exosomes into a single customizable pipeline.

The significance of the launch lies in the persistent challenge of solid tumors. While CAR-T therapy has transformed outcomes in hematological malignancies, its impact on solid tumors remains limited by three fundamental obstacles: poor T-cell infiltration through dense tumor stroma, an immunosuppressive tumor microenvironment (TME) that drives metabolic exhaustion, and safety concerns such as cytokine release syndrome (CRS), along with antigen heterogeneity and escape. Conventional CAR-T approaches alone have struggled to surmount these barriers simultaneously, according to the company.

Creative Biolabs' platform seeks to address each bottleneck through distinct vesicle functions. The OMV engineering arm functionally remodels the tumor microenvironment to promote CAR-T infiltration. Engineered OMVs can carry selected immunomodulatory or delivery payloads for investigating tumor-microenvironment remodeling and CAR-T infiltration. A standout innovation is the BROAD-CAR system, in which OMVs transiently program tumor cells in vivo to display synthetic antigens recognized by a universal CAR-T construct, providing a preclinical strategy for evaluating antigen redirection across heterogeneous tumor models.

Additionally, OMV surface engineering with targeting ligands guides CAR-T homing precision, while detoxified LPS strains require characterization of immunostimulatory activity and endotoxin-related safety, a critical differentiator from synthetic liposome carriers that lack intrinsic immune-activating capacity.

The exosome engineering arm provides a biocompatible support system to sustain CAR-T activity within the suppressive TME. CAR-exosomes, which directly display functional CARs on their surface, deliver antigen-specific tumor killing without live cell administration, with a safety profile that requires preclinical comparison with live-cell CAR-T approaches. Selected exosome systems are being investigated for transport across biological barriers, including the blood-brain barrier, and can be engineered to carry cytokines, checkpoint inhibitors, and survival signals intended to modulate CAR-T exhaustion and persistence.

A third service line equips both vesicle types with customized therapeutic payloads. Small molecules, nucleic acids such as mRNA, siRNA, and miRNA, and proteins can be co-encapsulated within a single vesicle for multimodal synergy. Metabolic TME remodeling formulations alleviate iron-mediated oxidative stress, while vesicle-mediated checkpoint blockade enables localized, sustained PD-1/PD-L1 inhibition directly at tumor sites, maximizing activation while minimizing systemic toxicity.

The company said researchers evaluating vesicle-engineering partners should assess dual-modality integration, LPS detoxification data, and cargo co-loading capacity. The platform's differentiators include antigen-independent targeting via synthetic marker installation on cancer cells, payload capacity and immunogenicity characterized relative to selected synthetic carriers, cell-free CAR-exosome options for preclinical repeat-dosing and formulation studies, project-specific ex vivo workflows designed to preserve favorable T-cell phenotypes, and full technical validation including NTA sizing, high-resolution imaging, and pharmacokinetic profiling.

For the research community, the launch matters because it offers a dual-mechanism toolkit intended to investigate stroma penetration and metabolic persistence in parallel rather than sequentially. A senior scientist at Creative Biolabs said the field has accepted that solid tumors require a fundamentally different approach to CAR-T therapy, and that integrating OMV and exosome engineering into a single platform gives researchers a way to investigate these barriers in parallel, from stroma penetration to metabolic persistence.

Researchers can access the full technical brief on OMV-exosome CAR-T engineering through the company's CAR-T development services page. The platform is positioned for preclinical solid-tumor research, where the combination of vesicle-mediated remodeling and payload delivery could influence how laboratories design next-generation CAR-T strategies.
